Scotty, my 2019 Camry with the 8 speed automatic transmission has that mode where I can manually shift it and it has the paddle shifters. Will having it in the S +/- mode and then shifting it back to “Drive” while the car is moving harm the transmission?

No it will not. Switching it back to D while driving will just shift to the optimal gear for fuel efficiency.
No, won't hurt it at all if that's how you want to drive it. It doesn't make up for the absence of a manual. Indy cars and F1 cars use paddle shifters, so several years ago some manufacturers started putting the paddle shifters in and then other's have joined in. IMHO, it's a gimmick to give the cars a little more sporty feel to 'em. I had it in several cars; didn't like it, just put in drive and go.
