My 2000 Toyota 4Runner with 205,000 miles recently vibrated and won’t go over 40 miles/hour then engine light came on P0303 and p0171. When I let the truck sit for a while and start it again then it run fine. One way to trigger the misfire is to put it in reverse. I have replaced the spark plug and coil on cylinder 3. I don’t hear or see vacuum leak. The problem still exist. Please help. Could it be fuel injector on cylinder 3 or electrical short or something else. Thanks

Misfire is covered in our frequently asked questions (FAQ).
Try swapping the injectors.

I had the same sort of issue and turned out it was my fuel pump.
fuel pump typically affects all cylinders (P0300). He has misfire in cylinder 3 only
