We have a 04 Toyota Sienna with 243k miles. It has all new brake pads/rotors and tires all the way around. It has a prominent shake at highway speed. Where should I look first.

Well assuming that the brake job was done correctly, Out of balance tires is a common issue that can cause a shake at high speeds, I'd look into that first. If the shaking only happened after these changes that is.
It did shake before new tires. That’s was my first thought.
