Can a tire really go flat after inflating with air?

Yes, if the hole in the tire is not sealed.

Certainly if something from the road punctured the tire or there's some other defect like a bad tire valve or bead not seating properly on the rim.

Like @chucktobias said, check your valve stem, make sure it's threaded all the way in and seated. Squirt some soapy water on it to see if it bubbles.
