Vehicle has 49K miles, garage kept, 2nd owner. If it checks out after 200 mile quick trip to see if it burns any oil and a mechanic says OK, I am thinking of offering $4500 to $5000.
Would this be good buy? Any issues with 09 Honda Accord EXL 6s.

Which transmission (manual or automatic)?
They are well made cars. If I were you I would have a mechanic check it out as well. Besides the engine oil burning (mostly on engines equipped with VCM, @daywalker probably knows which engine it is), the V6 also tends to have excessive brake wear, check for that. They also tend to have uncomfortable seats that becomes evident after around 20 minutes, you will probably notice it or not notice it after your 200 mile test drive.

To be fair, it was a pretty problematic model year as it was the second year of the generation. I think the oil burning was mainly for the 4 cylinder engine, but I might be wrong. Safe to say you should avoid the engine with cylinder deactivation, or VCM, and like you said check for oil consumption. The other two things to look out for are the uncomfortable seats (which you would quickly see if its fits you or not) and the excessive brake wear.
49k miles is basically NOTHING if its the real miles. Get your hands on service records. Make sure the oil was changed every 5k miles and the transmission fluid was drained and filled (not flushed) if not you drain and fill it if you buy it... Maintenance is key. If it checked out, $5000 sounds like a great price.
If it is a manual transmission, then the V6 does NOT have cylinder deactivation (what Honda calls VCM). However, if it does have an automatic transmission then it does have VCM. For longevity, I would avoid engines with cylinder deactivation as it can potentially wear out/damage the engine over time and just another layer of complexity that can give issues later.
