Would you check the automatic transmission fluid for a 2010 toyota corolla when the engine is running or when its off and hot?

If you are talking about trans level, then for most cars you want the engine running and hot before you check the dip stick if your car has a dipstick. If you check it when the engine is off you may get a false reading. Some cars have a nice "hot" and "cold" checks on the dipstick as well.
