I recently bought a 2022 4Runner TRD Off Road Premium with 28k miles. I went to a concert and the dust from the ground went into the door frame where the window goes up/down. When I try to roll down the windows, the windows screech and sound like they will break. Any recommendations?

Clean up the window track and seal, and spray some silicone lube down the track. If that doesn't do it you'll need to get inside the door to do a more thorough cleaning.
