I have a 2016 Toyota 4Runner Limited with 101,000 miles, automatic transmission. I don't have the maintenance history as I bought it a couple months ago from a dealer. My mechanic recommends Amsoil synthetic oil, should I switch to this synthetic in your opinion, any benefits over OEM oil? Should I have the transmission fluid replaced or is the mileage too high? I am having no problems, runs like new. Thank you.

as far as I know, OEM's switched to synthetic transmission fluid by the early 2000's
Dexron VI is synthetic, and I'm pretty sure Toyota WS is too.

If you like over paying for transmission fluid go ahead and use the Amsoil. There are plenty of other trans fluids that are just as good at substantially less cost.
Nothing wrong with Amsoil but the price. Probably a good idea to change trans fluid now as long as the fluid is not burnt and gritty.

Amsoil synthetic is expensive. I personally use oem oil filter which I buy from the dealership when they have a sale for buying 3 or more. And then use Kirkland Signature motor oil from Costco.
My bad, for some reason I read the question as for motor oil. As for the fluid I just use the OEM one again buying it when on sale. And also for my Honda for example the owners manual explicitly says to use the OEM fluid.
I was referring to engine oil. I was also wondering if I should change transmission fluid, but thought I has read with over 100,000 miles maybe I should not. Thank you.
