Hi,
Why doesn't my key lock or unlock the driver's door cylinder on my 2008 Lexus RX 350 ?

Either the key or the lock cylinder is worn out.

I’m assuming it wasn’t used much, Have you tried WD-40? And just jiggling it around? (At your own risk 🤪)
On my Toyota not all keys that work in the door worked on the truck and it was just a matter of just playing around with the key until the cylinder began accepting it.
(the key gets worn out together with the cylinders it’s used in, it can get so worn that it’s be beyond the (not worn out yet) cylinder’s tolerances)
