I have a 2006 Ford Mustang V6 with 81k and have noticed a leak that I think is from the water pump weep hole or water pump gasket. If I pressurized the system will it leak through the weep hole to show an issue? I know it will leak if the gasket is bad.

Yes, it will leak under pressure, that's what the weep hole is for. When the bearings go bad, rather than going until catastrophic failure, the bearing wear allows fluid to seep past them, and out the weep hole. The thought is you will notice antifreeze on the ground and replace the water pump.
Hook up a cooling system pressure gauge and pressurize the system to the rating on the radiator cap. It will bleed pressure and leak.
