Brake fluid bleeding question

Merry Christmas everyone !

2019 Subaru Outback 2.5i

Looking to do Brake Fluid Replacement.

After attaching the vinyl tube to the bleeder and the other end into a bottle with some brake fluid:

Is it a must to tighten the bleeder screw after every press of the brake pedal ?

Or is it ok to loosen the bleeder, press-release the brake pedal multiple times until clean fluid comes out and tighten the bleeder back at the end ?

You have to tighten the bleeder screw after each press of the brake pedal to draw the new fluid into the system.  This is why it takes two people, or, if you have a Mighty Vac, you can do it yourself.
