Is it necessary to have a car running and up to temp. to check the coolant with a multimeter? Some videos tell you to do that and others don't.

well if its hot & you open the cap, it'll spray everywhere and burn you. Do not open it if its up to temperature. Only take off the radiator cap if the engine is completely cool

Check it cold using one of these.
And change it at least every 5 years.

I'd only do that if it's sat for a while, and would run it to operating temp, then let it cool completely to get everything moved around. If you drove it yesterday, there's no real reason for the stuff in the radiator to not be uniform with the antifreeze in the block and heater core.
