as we know. coolant is use for anti freezing agent as well as to cool down engine block... is there any direct effect of fuel efficiency if we cool down engine using coolant?

Engines are designed to operate ideally at a certain temp, hence thermostats. In the old days the cooler it was the better but today with all the sensor feedback controls, they need to operate at the temp designed for.

Yes. The engine has an optimal temperature range where it is most fuel efficient. The cooling system is designed to keep the engine in that range.
