What should I check first, second and third , etc ? Help !

It might be helpful if you provided the actual code(s).
Anything and everything that can interfere with the combustion process can potentially cause a misfire. Low hanging fruit would be things like spark plugs, coils, fuel and air filters, vacuum leaks. However more diagnosis is required - guessing and throwing parts at a problem rarely produces a good result.

So it only drives 1,200 miles per year. Not much. Cars don't like long periods of sitting and neither does fuel. It tends to gum things up.
I would be looking at fuel related problems. Clogged injectors, fuel filter, etc. First thing I would try is some fuel injector cleaner. In the meantime, find a gauge to measure fuel pressure.
Code P.O. 316.
It's not PO as in PO Box. It's a 'P' for Powertrain, and then a 4 digit numerical code. (in case you're doing research online)
DTC P0316 definition is "Engine Misfire at Startup".

I'm not sure where you got the idea that it's cylinder #1 misfiring. Were there other codes?

it was a vacuum tube leak I just taped it up
if it leaks in one spot, it's going to leak everywhere. It's a 21 year old car and these things just break down. Replace the whole thing. Vacuum hose is very inexpensive.

Thank you Chuck, now, could you tell me how to erase the original post? By the way I fixed the problem, it was a vacuum tube leak I just taped it up and Erased the code. Never came back after I erased it, and the misfiring engine worked as smooth as the day it was bought. Thanks everyone for your suggestions
