I bought this van over 5 years ago with 140k miles on it. It's about to hit 210k, and I've done nothing but regular oil changes (whenever the required maintenance light would come one), brake pads, and tires. Oh, and I paid a local shop to change the A/C compressor that went out 2 or 3 summers ago. I didn't know about Toyota's longevity (this one was made in Indiana) when I bought it, and thought I'd do whatever to keep it running. Now, I'm going to get more serious about it. Going to change fluids and spark plugs.
#1 Should I change out the transmission fluid? I've noticed in the past, it occasionally hesitates before shifting, but nothing more than that.
#2 What else should I consider as I now want this to be the last mini-van I ever buy?
#3 (bonus round time) The passenger side automatic sliding door stops working every January, and starts back working before March. Bought the thing in 2016 (which is probably why the family sold it in January) and it's done it every year. And part 2, just recently, the driver's side sliding door, when I park on a hill, sometimes won't lock in the open position and will slide closed. I can push it open and it will lock. Any thoughts?

If you've never changed the trans fluid, I wouldn't start now. A change every 5 years/60k miles is the recommendation so if that hasn't been done I'd leave it and get as much as you can out of it.
All you can do really is keep up with the recommended maintenance. That will get you the best longevity possible.
As for the doors, it sounds like an electrical short. Probably will need a mechanic to track that one down and fix it as there are miles of wiring in those vans.
