Scotty I agree with you about RFT tires however my Toyota Sienna only comes with them so there is no choice.

Yes you can. Just need to decide on a contingency to deal with a flat tire situation. Get a spare tire or small compressor and a can of fix-a-flat, etc. AAA is a good idea too.
Commonly done on cars in the 1960s that came with run-flat tires, like 3-seat Rambler station wagons where there was no room for a spare tire. I don't think anyone ever bothered installing new run-flats once the originals needed to be replaced. As others have mentioned you just need to be equipped for a flat tire situation whether that's going to be AAA (they may takes hours to get to you), carrying repair plugs and a 12V compressor, or just tossing a spare tire in wherever it can fit.
