Should I buy a 2000 Chevrolet S10 regular cab long box truck with a 4.3L V6 automatic transmission with 153,000 miles on it for $3500?
A definite "maybe". The way pickup prices are distorted these days, the S10 probably gives you about as much bang-for-the-buck as there is. You certainly get more for your money than an old Tacoma (which people are really over paying for right now, even accounting for reliability).
Problem is they are all getting old, and frankly weren't designed to get old. The parts are not particularly robust. Performance is not great either. Or looks for that matter, particularly for the second gen models, which are homely and unloved (the older square body style is appreciating a bit).
However, the power trains, including both 4- and 6-cylinder are pretty good. They are also really cheap to work on. And if you just need a little grocery-getter that can maybe haul your fishing poles to the lake, they are perfectly fine.
They are getting to be fewer, but are by no means rare. So if you want an S10, don't settle for a bad one. Make sure the motor and transmission is inspected before you take it home. And I'll second @gew above to verify that there is no serious rust issue.

PS: my brother inherited by Dad's 97 S10 and is restoring it as we speak. When I checked it out myself a couple years ago, it seemed to be mechanically sound even after all these years.
No. That's the price I paid 16 years ago. With less mileage.
