I have a 2012 Ram 1500 4.7L V8 (flex fuel capable). I have never used E85 in the vehicle and was wondering if switching to it after 10 years would mess up anything?

It is possible. I would not risk it especially considering the fact that it does not financially make sense using E85 instead of E10.
I've been using Regular 87 octane for the past 10 years and when gas prices went up, I considered trying to use E85.
I've heard many things like E85 isn't good if your car sits for awhile. My daily commuter is a motorcycle so I've gone as long as 2 weeks to a month without driving my truck.
I've also heard that E85 isn't good for the rubber seals. But I don't know.
I'm not sure about E85 effect on the fuel system if the truck sits for a long time but I remember Scotty always mentioned that if you have not used E85, your better not to use it.

Flex fuel vehicles use compatible materials.
The ethanol added to gasoline is "hydroscopic", that is it absorbs water from the atmosphere. This is a problem with the 10% ethanol gas, if it sits for a while. I would imagine it would be a major problem in a vehicle that was only used a couple times a month. I'd use a fuel stabilizer, or better yet, stick with regular gas.
The word meaning tendency to absorb moisture is "hygroscopic" with a 'g'.
